Abstract
The primary tool for analyzing oligosaccharide conformational heterogeneity is 1H nuclear magnetic resonance (NMR) spectroscopy, which utilizes nuclear Overhauser effect (NOE) measurements to generate interatomic distances used to predict a range of three-dimensional (3D) structures for oligosaccharides. Oligosaccharide flexibility influences the avidity of carbohydrate-protein interactions by allowing repositioning of terminal sugar residues into a three-dimensional array that is complementary to the binding site topography on carbohydrate receptors. It is primarily the underlying oligosaccharide “scaffolding” which folds at one or more of the glycosidic linkages, resulting in multiple configurations of terminal sugar residues, only one of which is recognized by the receptor.
