Abstract
This chapter describes the immunochemical assays for toxic shock syndrome toxin-1 (TSST-1). The chapter discusses the assay to assess the capacity of tampons to alter TSST-1 production. (TSST-1) is a low-molecular-weight single peptide toxin secreted by some strains of Staphylococcus aureus. The toxin is highly associated with menstrually related toxic shock syndrome (TSS) and is considered as a causative toxin. An assay was developed based upon double immunodiffusion to quantify TSST-1 for the purpose of elucidating both conditions that yield high levels of toxin for ease of purification and the possible mechanism of tampon involvement in TSS. TSST-l-producing S. aureus are cultured in the desired medium, usually from 1 to 100 ml. Subsequently, 4 vol 4° absolute ethanol is added (it is not necessary to remove the cells), and the cultures placed at 4° for at least 1 hr to allow the TSST-1 to precipitate. The sensitivity of the assay may be increased 2- to 4-fold by drying and staining the slides.
