Abstract
The fluorinated pyrimidines 5-fluorouracil (FUra) and 5-fluoro-2′-deoxyuridine (FdUrd) are drugs that have been shown to have clinical activity against colorectal carcinoma and other malignancies. Attempts to enhance the effectiveness of FUra have been based on considerations of both the biochemical pathways of FUra activation and the site of FUra action, which in some cases involves inhibition of thymidylate synthase (TS) by 5-fluorodeoxyuridine monophosphate (FdUMP), the active metabolite of both FUra and FdUrd and in others involve the incorporation of 5-fluorouridine triphosphate (FUTP) into RNA.1–4
