Chromatin Immunoprecipitation in Aspergillus fumigatus

Abstract
Aspergillus fumigatus is the major human filamentous fungal pathogen. While its importance as a pathogen is well-appreciated, our understanding of its biology is still at an early stage. One valuable area of study has been the analysis of gene expression in A. fumigatus. Dissection of the transcriptional circuits that control drug resistance and virulence in this fungus has been facilitated by the use of chromatin immunoprecipitation (ChIP), an approach pioneered in the mid-1980s. The use of ChIP coupled with next-generation sequencing (ChIP-seq) allows the determination of the genomic binding sites of a transcription factor of interest. Here, we describe the application of ChIP in A. fumigatus and detail different approaches to appropriately shear chromatin from this organism. This sheared chromatin can be used for analysis of factor association with single genes by qPCR or for downstream ChIP-seq applications.Aspergillus fumigatus is the major human filamentous fungal pathogen.
