Abstract
Alzheimer's disease (AD) is increasingly becoming a significant public health concern, affecting over 6 million Americans by the end of 2021. Metabolomics, the study of metabolites on a systemic scale, offers valuable insights into cellular processes and holds promise for identifying biomarkers related to AD. This study analyzed a metabolomics dataset consisting of 145 samples, including 76 “pure” control samples and 18 “converter” control samples, to identify significant biomarkers associated with Alzheimer's disease. Initial data analysis involved unsupervised learning to evaluate metabolomics data, followed by differential analysis to identify significant metabolites, with further screening using Lasso regression. Gene annotation of microRNA data and normalization of gene expression were performed, and PCA was used to select the paired MCI/AD converter group. Differential expression analysis, c1onducted using the Limma package in R, identified 13 metabolites showing significant concentration differences between the "MCI/AD" and "converter" groups. A Lasso regression model, built using these metabolites, highlighted two key metabolites, 122.0969_9 .6283 and 235.0928_2.7431, as significant biomarkers for Alzheimer's disease. These findings suggest that these metabolites could be involved in the initiation and progression of AD, presenting potential as prognostic markers or therapeutic targets for the disease.
