Normal mammary gland development is a complex process that involves cellular proliferation and differentiation. Disruption of this process can initiate tumor development. However, a better understanding of the molecular mechanisms and factors that regulate mammary gland development and breast cancer tumorigenesis is needed. Transcription factor AP-2ɣ is a regulatory protein that has been implicated in the control of proliferation, differentiation, and apoptosis pathways. HER2-enriched breast cancer has traditionally been associated with an aggressive disease course and has been shown to correlate with AP-2ɣ levels. The aim of this thesis is to provide a comprehensive analysis of the regulatory role of AP-2ɣ gene targets and pathways in mammary gland development and breast cancer initiation and growth. We will use various genetically modified mice, cultured cancer cells, and sequencing analysis of normal mammary gland as well as breast carcinomas to study the factors involved in mammary development and breast cancer.
