Abstract
Craniofacial disorders result from a disruption of normal embryogenesis of the head and can be caused by genetic, environmental and sporadic events. Isolated clefts of the lip and/or palate are common such defects and this review will summarize what is known about their common Mendelian and environmental triggers. Further in the last few years we have begun to identify the common genetic contributors to isolated forms of cleft lip/palate. Recent findings from DNA sequencing and genome-wide association will be summarized for these loci that have been reliably confirmed.
