Abstract
The synthesis of glycine-labeled collagen can be followed by a sensitive assay that detects radioactivity in glycine phenylthiohydantoin produced from
N-glycyl peptides generated by clostridial collagenase. The assay is sensitive and highly specific for the direct measurement of collagen synthesis. Many commonly used assays are indirect, in that they measure the difference in TCA-precipitable radio activity before and after collagenase treatment. In addition, the new assay is less affected by nonspecific proteases, which contaminate many commercial collagenase preparations and lead to erroncously high estimates of collagen by the indirect assay.
