Aldosterone (ALDO)‐Salt induced activation of the Lamina Terminalis (LT) and paraventricular nucleus (PVN) neurons that express mineralocorticoid receptors (MR) in rats

Abstract
Studies from our laboratory have shown that 28 days of ALDO infusion induces a significant increase in blood pressure. This response requires the activation of central MR. The neuronal pathways activated during ALDO‐Salt induced hypertension are not well established. The purpose of the present study was to test the hypothesis that ALDO‐Salt treatments results in activation of MR expressing neurons in the LT and PVN, which project to the sympathetic controlling area, rostroventrolateral medulla (RVLM). The retrograde tracer fluorogold (FG) was iontophoretically delivered into the RVLM of male rats. Aldosterone was administration (0.75 μg/h, for 28 days, sc) with 1% NaCl as the only choice of drinking solution. Control animals received vehicle and water ad libitum. On the day 28, the animals were perfused with 4% paraformaldehyde for brain fixation and further immunodetection of Fra‐like proteins, as markers of neuronal activation, MR and FG. ALDO‐Salt rats expressed Fra‐like in the cells of the organum vasculosum of the lamina terminalis (OVLT), median preoptic nucleus (MnPO) and PVN, but not in the subfornical organ (SFO). In the PVN, Fra‐like was notably co‐localized with MR and FG. These findings suggest that neurons of the LT and PVN are activated by ALDO‐Salt treatment, and that ALDO‐salt activates PVN neurons that directly project to the RVLM, probably by acting via MR.
