CO2 utilization and distribution during H+ secretion by bullfrog fundic mucosa

Abstract
Spontaneously secreting in vitro sheets of bullfrog fundic mucosa deprived of exogenous COz were treated with known stimulants and inhibitors of H’ secretion. Changes in secretion correlated closely with the CO2 distribution across the mucosa, where ACO, equals the difference between nutrient and secretory COz. Stimulation with histamine plus theophylline produced equimolar increases in H’ secretion (i.28 & 0.25 to 2.10 t 0.27 peqocrnD2* h-l, P < 0.001) and AC02 (1.46 & 0.23 to 2.3 * 0.35 peq*cmB2* h-l, P < 0.01) associated with an increase in total CO2 production (3.0 & 0.27 to 3.8 & 0.46 peg* crnD2* h-l, P < 0.01). Complete inhibition by anoxia (100% N2) produced equimolar reductions in H+ (1.49 k 0.16 to 0 peq* cms2 l h-l, P < 0.001) and ACO, (1.6 t 0.10 to 0 peq. crna2* h-l, P < 0.001) associated with near-total cessation of oxidative metabolism, as reflected by the decrease in total CO2 production (3.72 t 0.35 to 0.96 k 0.07 peqocm-20 h-l, P < 0.01). Partial inhibition of secretion (by substituting a chloride-free solution, except for Cl- in the titrant) on the nutrient surface produced equimolar decrements in H+ (1.30 t 0.17 to 0.76 t 0.03 peq. cm-** h-l, P < 0.05) and AC02 (1.3 t 0.22 to 0.68 t 0.16 peg0 crns2. h-l, P < 0.01) without influencing oxidative metabolism or total CO2 (3.02 t 0.25 to 2.92 t 0.26 peqocm-2 l h“, NS). A model is proposed for CO2 distribution as a method for distinguishing between the various causes of decreased titratable H+ on the secretory surface of in vitro fundic mucosal sheets. Additional information regarding the metabolic behavior of the oxyntic cell and the phenomena associated with H+ secretion may be obtained by this method.
