Inhibition of calcium‐independent phospholipase A2 in pulmonary microvascular endothelium prevents inflammatory mediator production

Abstract
Mast cell tryptase release increases calcium‐independent phospholipase A2 (iPLA2), activity in human pulmonary microvascular endothelial cells (HPMEC). The barrier formed by these cells between external pathogens and the bloodstream can be compromised by the subsequent increase in production of membrane phospholipid‐derived inflammatory mediators and increased expression of cell surface adhesion molecules that can recruit inflammatory cells from the bloodstream. Our results demonstrate that tryptase stimulation of HPMEC increases iPLA2 activity (1.6 fold), arachidonic acid release (3.5 fold) , prostacyclin (PGI2) release (2.6 fold), and cell surface expression of P‐ and E‐selectin that is inhibited by pretreatment with bromoenol lactone (BEL, a selective iPLA2 inhibitor). Up‐regulation of cell surface adhesion molecules can lead to increased recruitment of inflammatory cells from the blood stream including mast cell precursors and neutrophils. In conclusion pharmacological inhibition of iPLA2 may provide a therapeutic avenue to reduce the recruitment of inflammatory cells to areas of inflammation in the lung and thus interrupt or delay the pathogenesis of asthma..
