Preparation of urea and guanidinium chloride stock solutions for measuring denaturant-induced unfolding curves

Abstract
The preparation of the chemical denaturant used in generating protein unfolding curves must be done with great care. Prepare urea stock solutions by weight and check the concentration by refractive index measurements. Guanidinium chloride is quite hygroscopic, and thus it is difficult to prepare stock solutions by weight. Base the molarity of guanidinium chloride stock solutions on refractive index measurements. This protocol describes the preparation of approximately 100 ml of an approximately 10 M urea stock solution containing 30 mM MOPS buffer (pH 7.0). A guanidinium chloride stock solution is prepared in the same manner, merely substituting the appropriate equations as stated in the text.
