The effects of L‐NAME and Tempol in Ang II induced hypertension in male mice

Abstract
It has been shown that superoxide is involved in ANG II‐stimulated activation of the central nervous system (CNS), and that nitric oxide (NO) acts as a sympathoinhibitory substance within the CNS. The present study tested the effects of L‐NAME and Tempol on Ang II induced hypertension. Aortic blood pressure was measured in conscious, freely moving wild‐type male mice with the use of telemetry implants. Tempol (50 μg/kg/min, n=7), L‐NAME (500 μg/kg/d, n=3), or both (n=5) were chronically infused into lateral ventricle by brain infusion assembly with attached osmotic pump. ANG II (800 ng/kg/min) infused subcutaneously via an implanted osmotic pump increased MAP 31.1±2.4 mmHg (107.8±1.1 to137.8 ± 1.8 mmHg, n=6). Central infusions of Tempol, L‐NAME, or both did not change the basal blood pressure. However, Tempol attenuated the pressor effects of Ang II in male mice (137.8 ± 1.8, n=6 to 114.4 ± 2.6 mmHg, n=7), while L‐NAME had no effect (137.8 ± 1.8, n=6 to 133.3 ± 1.9 mmHg n=3). The change in blood pressure induced by Ang II during central infusion of both Tempol and L‐NAME was greater than that seen in infusion of Tempol alone (18.6 ±2.5, n=5 vs 10.9 ± 2.3 mmHg, n=3). Together, these data indicate that superoxide is a key mediator of excitatory actions of Ang II in CNS and that NO may modulate these effects.
